Metagenómica dirigida al gen 23S ADNr para la identificación de microorganismos asociados al estómago de Anadara tuberculosa (Sowerby, 1833), agua y sedimento del manglar de Tumbes, Perú.

Resumen
La concha negra (Anadara tuberculosa) es una especie simbólica del ecosistema manglar, cuyas poblaciones están dramáticamente declinando debido a la sobrepesca y destrucción de su hábitat. Las iniciativas de producción de este bivalvo en laboratorio, han evidenciado dificultades que requieren extender nuestros conocimientos sobre su ecofisiología en su ambiente natural. En este trabajo se buscó caracterizar los microorganismos asociados al estómago de A. tuberculosa, agua y sedimento, obtenidos a diferentes salinidades del manglar de Tumbes – Perú, por metagenómica dirigida al gen 23S ADNr plastídico mediante secuenciación de próxima generación (NGS). Como parte de los resultados se logró extraer ADN total de las muestras de estudio de tres estaciones (9 muestras en total) y amplificar por PCR convencional la región del gen de interés, cuyo tamaño de los productos amplificados fueron de 450 pb. Posteriormente, de la secuenciación metagenómica, se obtuvieron un total de 588,200 secuencias, las cuales en su mayoría no fueron clasificadas a OTU’s. De las secuencias clasificadas, corresponden principalmente al dominio Bacteria (>50%) y Eucariota (<50%). El índice de Shannon-Weaver sugieren una alta diversidad para las muestras de agua, seguidas por la muestras de sedimento y con menos diversidad las muestras de estómago. Esta diversidad estuvo representada a nivel de filo por, Ascomycota, Proteobacteria, Firmicutes, Bacteroidetes, Cyanobacteria, Spirochaetes, Chlorophyta y Euryarchaeota como lo más representativos. Además ubicamos a organismos pertenecientes a los géneros de Metarhizium, Bacillus, Lactobacillus, Pseudomonas, Trypanosoma, Novosphingobium, Rhodopirellula, Oceanimonas, Rhodotorula, Volvox, Oscillatoria, Morganella, Chlorella, Shewanella entre otros. Estos resultados encontrados tienen un alto potencial para reorientar el proceso de cultivo de A. tuberculosa en base a su alimentación con el uso de probióticos y el cultivo de microalgas que aún no han sido consideradas en su dieta. Además de que permiten tener una visión más real de lo que sucede con la especie y su entorno.
Anadara tuberculosa is a symbolic species of the mangrove ecosystem, whose populations are dramatically declining due to overfishing and destruction of its habitat. The production initiatives of this bivalve in the laboratory, have shown difficulties that require extending our knowledge about their ecophysiology in their natural environment. In this work we sought to characterize the microorganisms associated to the stomach of A. tuberculosa, water and sediment, obtained at different salinities of the Tumbes mangrove - Peru, by metagenomic directed to the 23S rDNA plastid gene by next generation sequencing (NGS). As part of the results, it was possible to extract total DNA from the study samples from three stations (9 samples in total) and amplify the region of the gene of interest by conventional PCR, whose size of the amplified products was 450 bp. Subsequently, from metagenomic sequencing, a total of 588,200 sequences were obtained, which were mostly not classified to OTU's. Of the classified sequences, they correspond mainly to the domain Bacteria (> 50%) and Eukaryote (<50%). The Shannon-Weaver index suggests a high diversity for the water samples, followed by the sediment samples and with less diversity the stomach samples. This diversity was represented at the phylum level by Ascomycota, Proteobacteria, Firmicutes, Bacteroidetes, Cyanobacteria, Spirochaetes, Chlorophyta, and Euryarchaeota as the most representative. We also locate organisms belonging to the genera of Metarhizium, Bacillus, Lactobacillus, Pseudomonas, Trypanosoma, Novosphingobium, Rhodopirellula, Oceanimonas, Rhodotorula, Volvox, Oscillatoria, Morganella, Chlorella, Shewanella and others. These results have a high potential to reorient the process of cultivation of A. tuberculosa based on their diet with the use of probiotics and the cultivation of microalgae that have not yet been considered in their diet. Besides that they allow to have a more real vision of what happens with the species and its environment.
